Application Notes:
The D33 antibody is suitable for the detection of desmin by Western blotting and detects a band of 53kDa in muscle tissue extracts. The antibody is also suitable for the detection of desmin by immunocytochemistry on permeabilized cells, and Immunohistochemistry on frozen and formalin fixed, paraffin embedded tissues. The antibody works on paraffin-embedded material without pretreament with proteolytic enzymes, but the section should be pre-treated using heat mediated antigen retrieval with Tris-buffer containing 1 mM EDTA, pH 9,0 for 20 mins. Optimal antibody dilutions for the different applications should be determined by titration.
