Purified splenic NK cells from the LEW rat strain.
Natural killer (NK) cells are lymphocytes that mediate cytotoxicity and secrete cytokines after immune stimulation. Several genes of the C-type lectin superfamily, including the rodent NKRP1 family of glycoproteins, are expressed by NK cells and may be involved in the regulation of NK cell function. The KLRB1 protein contains an extracellular domain with several motifs characteristic of C-type lectins, a transmembrane domain, and a cytoplasmic domain. The KLRB1 protein, NKR-P1A or D161, is classified as a type II membrane protein because it has an external C terminus. NKR-P1A, the receptor encoded by the KLRB1 gene, recognizes Lectin Like Transcript-1 (LLT1) as a functional ligand. Human CD161 is a homodimeric disulfide-linked type II membrane bound glycoprotein found mainly on NK cells. CD161 has been implicated in the regulation of NK cell mediated cytotoxicity. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Optimal dilution determined by the researcher. Recommended Negative Controls: I1904-78X: IgG1 Murine Negative Control (For rat tissue) (FITC) Hybridoma: Spleen cells from immunized BALB/c mice were fused with cells of the mouse X63.Ag8653 myeloma cell line. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
