Human CD166 is a type 1 glycoprotein expressed on activated T cells, B cells and monocytes and appears to be the ligand for CD6 (1,2). Human CD166 may be important for activation of T cells. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry and as a binding blocker of CD6 to CD166. Other applications not tested. Recommended Dilutions: Flow Cytometry: 5ug/ml Immunohistochemistry: frozen sections Optimal dilutions to be determined by the researcher. FACS Protocol: 1. Five x 10e5 Cultured Raji cells per tube were washed and preincubated 5 minutes with 20ul of 250ug/ml human IgG (to block non-specific binding). 2. After which, cells were incubated 45 minutes on ice with 80ul of C2548-90M3 at 5ug/ml. 3. Cells were then washed twice and incubated with IgG (FITC) goat x mouse 4. Cells were washed three times, fixed and analyzed using FACS. 5. Cells stained with a mean shift of 0.87 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
