Human CD166 is a type 1 glycoprotein expressed on activated T cells, B cells and monocytes and appears to be the ligand for CD6 (1,2). Human CD166 may be important for activation of T cells. Applications: Suitable for use in Flow Cytometry and blocking of binding of CD6 to CD166. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ug/ml. 80ul labels 5x10e5 human Raji cells Optimal dilutions to be determined by the researcher. Protocol: 1. Five x 10e5 cultured human Raji cells were washed and incubated 45 minutes on ice with 80ul of C2548-90M4 at 10ug/ml. 2. Cells were washed twice and incubated with secondary reagent S7973-88P, Streptavidin(PE). 3. Cells were washed three times, fixed and analyzed by FACS. 4. Cells stained positive with a mean shift of 1.49 log10 fluorescent units when compared to a Mouse IgG1 (Biotin) negative control at a similar concentration. 5. Binding was blocked when pre-incubated 10 minutes with 20ul of C2548-90M3 at 0.5mg/ml. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
