Human CD166 is a type 1 glycoprotein expressed on activated T cells, B cells and monocytes and appears to be the ligand for CD6 (1,2). Human CD166 may be important for activation of T cells. Applications: Suitable for use in Flow Cytometry and Antibody Blocking. Other applications not tested. Recommended Dilutions: Flow Cytometry: 1:50 (10ug/ml) Antibody Blocking: Blocks binding of CD6 to CD166. Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Human Raji cells were washed and incubated 45 minutes on ice with 80ul of C2548-90Mg at a dilution factor of 1:50 (10ug/ml). 2. Cells were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.29 log10 fluorescent units when compared to a Mouse IgG1/R-PE negative control at a similar concentration. 3. Binding was blocked when pre incubated 10 minutes with 20ul of 0.5mg/ml C2548-90M6 (anti-CD166 antibody). Storage and Stability: May be stored at 4C before opening. DO NOT FREEZE! Stable at 4C as an undiluted liquid. Dilute only prior to immediate use. Stable for at least 12 months at 4C. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. PE conjugates are sensitive to light.
