Alpha 2-Macroglobulin is a plasma protein mainly synthesized in the liver, which functions as a proteinase inhibitor. It is a tetrameric molecule with a molecular weight of 720kD, and usually occurs in serum at 1000-1500mg/L. Its main. Following conformational change, the proteinase is deposited within a central cavity where it is still active but prevented from further contact with its substrate. Removal of the complex is mediated by the generation of recognition sites which react with receptors on a variety of cells including macrophages and liver cells. Applications: Suitable for use in ELISA, Immunohistochemistry and Western Blot. Other applications have not been tested. Recommended Dilutions: ELISA: 1:20,000-1:100,000 Western Blot: 1:2000-1:10,000 Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
