PRL-3 Protein tyrosine kinase (PTKs) and protein tyrosine phosphatase (PTPs) play a fundamental role in regulating diverse cellular processes. One primary growth response gene, first identified in regenerating liver, encodes a novel 20kD nuclear PTP known as PRL-1 (phosphatase of regenerating liver). Other than the signature sequence for PTPs, PRL-1 is not homologous to either the dual specificity PTPs (cdc25 and MKP-1) or monospecific PTPs. Database searches using the PRL-1 sequence led to the identification of mouse PRL-2 and PRL-3, which exhibit 87% and 76% identity to mouse PRL-1 in their amino acid Sequence. All PRL proteins bear significant sequence homology to cdc14p and recently identified tumor suppressor PTEN/MMAC1. Northern blot analysis revealed that PRL-2 is preferentially expressed in skeletal muscle, while PRL-3 is preferentially expressed in smooth muscle, skeletal muscle and heart muscle. Both PRL-2 and PRL-3 are expressed at lower levels in other tissues. Human PRL-3 exhibits more than 95% identity to mouse PRL-3. It was identified in a cDNA library from an explanted human heart obtained from a patient with idiopathic cardiomyopathy It has been reported that PRL-3 plays a role in the modulation of intracellular calcium transients induced by angiotensin-II. It has also been shown that the PRL-3 gene may be related to colorectal cancer metastasis.
