Immunogen: Rat Spleen cell homogenate.
The CD163 cell surface glycoprotein is a 175kD molecule also known as ED2. CD163 is expressed by ~50% of peritoneal macrophages, a subset of splenic macrophages, and by macrophages in most other tissues. However, it is not expressed by monocytes, alveolar macrophages or microglial cells. May be used in histology using antigen retrieval, and has also been described reacting with paraffin-embedded material following PLP fixation (Periodate-lysine-paraformaldehyde). Applications: Suitable for use in Western Blot, Immunohistochemistry and Immunoprecipitation. Other applications have not been tested. Recommended Dilutions: Immunohistochemistry (Paraffin): Requires antigen retrieval by protein digestion (trypsin or pronase). Reported reaction following PLP fixation (Periodate-lysine-paraformaldehyde) Immunohistochemistry: frozen sections Optimal dilutions to be determined by the researcher. Positive Control for Immunohistochemistry: Liver Hybridoma: SP2/0-Ag 14 myeloma cells with spleen cells from BALB/c mice Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
