CD163 is a membrane glycoprotein of 130kD, which is expressed exclusively on the cell surface of human monocytes and macrophages that predominantly evolve in the late phase of inflammation. The CD163 is a member of the scavenger receptor cysteine-rich superfamily. Applications: Suitable for use in Western Blot, Flow Cytometry, ELISA, Immunoprecipitation, Immunofluorescence, Immunohistochemistry and Functional studies. Other applications not tested. Recommended Dilutions: Western Blot: 1:10. SDS-PAGE under non-denatured conditions. Block blot with 1% skimmed milk for 1h and incubate with C2548-90D2 (0.4ug/ml) in 0.1% BSA/TBS for 1h. Detection of ~130kD product in spleen extracted in presence of NP-40 or in glucocorticoid-treated human monocytes. (Ref 2,5) Flow Cytometry: 1:10. Permeabilization as well as no permeabilization is used. Block cells with 1% BSA for 30 minutes at 4C. (Ref 5) Immunohistochemistry (Frozen sections): 1:10 Functional Studies: Reduction of CD69 expression by adding 2.5ug/ml C2548-90D2 to 5x10e5 human T-lymphocytes in presence of exogenous CD163. (Ref 2) Immunofluorescence: Transfected CHO-cells were plated on coverslips and fixed in 4% paraformaldehyde for 20 minutes. Cells were stained with 4ug/ml C2548-90D2 for 60 minutes. Immunoprecipitation: 10ug C2548-90D2 was covalently coupled to beads (with 20 mM dimethyl pimelimidate), blocked with 0.2M glycine buffer for 2 hours and incubated with cell lysate for 2 hours (Ref 5). Optimal dilutions to be determined by the researcher. Recommended Positive Control: CHO-cells transfected with CD163 Recommended Negative Control: Muscle cells Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
