PTP1B, also known as PTPN1, is the founding member of the protein tyrosine phosphatase (PTP) family. PTP1B catalyze the hydrolysis of the phosphate monoesters specifically from tyrosine residues. PTP1B is involved in signaling pathways that regulate a variety of cellular processes including cell growth and oncogenic transformation and has been shown to act as a negative regulator of insulin signaling. PTP1B has also been reported to dephosphorylate epidermal growth factor receptor kinase, which implicates this phosphatase in cell growth control and cell response to interferon stimulation. Full-length recombinant protein corresponding to aa1-436 with an N-terminal GST-tag from human PTP1B, expressed in E.coli. Uniprot/Accession: P18031 Specific Activity: ~2,000 pmole/min/ug. One unit will hydrolyze 1nmol p-nitrophenyl phosphate per minute at pH 7.4 and 30C. Assay Buffer: 50mM HEPES, pH 7.4, 2mM EDTA, 3mM DTT, 100mM sodium chloride, 50mM pNPP. Applications: Suitable for use in enzyme kinetics, regulation, to dephosphorylate target substrates and for screening inhibitors. Other applications not tested. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
